The Giardia Lamblia (Feces) rapid cassette test is a rapid chromatographic immunoassay for the qualitative detection of Giardia Lamblia antigen in human stool samples. Parasitic infections remain a very serious health problem around the world. Giardia Lamblia is among the most common protozoa known to be a major cause of severe diarrhea in humans, particularly in immunocompromised people. Giardia Lamblia, also known as Giardia intestinalis, is a flagellated parasite that colonizes and reproduces in the small intestine, causing giardiasis. Parasitic infections remain a very serious health problem around the world. Giardia Lamblia is among the most common protozoa known to be a leading cause of severe diarrhea in humans, particularly in immunocompromised people. Giardia Lamblia, also known as Giardia intestinalis, is a flagellated parasite that colonizes and reproduces in the small intestine, causing giardiasis. The parasite attaches to the epithelium via a ventral adhesive disk and reproduces via binary fission. Epidemiological studies, in 1991, showed that Giardia infections increased in the United States with a prevalence of approximately 6% out of 178,000 samples. Generally, the disease goes through a short acute phase followed by a chronic phase. G. lamblia infection, in the acute phase, is the cause of watery diarrhea with elimination of trophozoites. The stool returns to normal, during the chronic phase, with transient emissions of cysts. The presence of the parasite on the wall of the duodenal epithelium is responsible for the malabsorption. The disappearance of the hairs and their atrophy lead to problems with the digestive process in the duodenum and jejunum, followed by weight loss and dehydration. However. most infections remain asymptomatic. Diagnosis of G. lamblia is performed under the microscope after flotation on zinc sulphate or by direct or indirect immunofluorescence, on unconcentrated samples viewed on a slide. The rapid test can detect Giardia Lamblia in fecal samples within 10 minutes. It is based on the detection of a 65 kDA coproantigen, a glycoprotein present in G. lamblia cysts and trophozoites. The Giardia Lamblia (Feci) rapid cassette test is based on a membrane technology that uses colloidal gold. A nitrocellulose membrane is sensitized with a direct anti-Giardia Lamblia antibody. The specificity of the test is guaranteed by an antibody specific for a Giardia Lamblia antigen conjugated with colloidal gold. This conjugate is dried on the membrane. The fecal sample must be diluted in the extraction buffer provided in the test kit. When the extracted sample comes into contact with the test strip, the conjugate migrates with the sample by passive diffusion and the conjugate along with the sample comes into contact with the anti-Giardia antibody in the test line T. If the sample contains antigen G lamblia, the conjugate-antigen complex will remain bound to the anti-Giardia reagent and a red line will appear. The solution will continue to migrate to encounter a second reagent which will bind to the control conjugate, thus producing a red control line which confirms that the test is working correctly. The result is visible within 10 minutes.
